Ace I hate to say this but there is not enough information you are providing.
Typically before you ever use spotlight to polypaint details from an image to your mesh, you will be done with zRemeshing and adding sub division levels. You would polypaint on your model at hi-res, perhaps at subdivision levels yielding as many as 1 million to 15 million polys or more. Polypainting is the latter part of your project. If you are mixing sub division levels and zRemeshing afer Polypainting you are going to get unpredictable results.
