I have installed ZBrush 4R2 today and it was running. I tried to restart ZBrush and it won´t start anymore.
It´s running on a Mac.
Do you get any error messages? Has anything changed about the system? (For example, if you’re on a network did your IT do some kind of network update last night?)
Have you tried reinstalling?

I have moved some files from the old installation (ZBrush 4) into Zbrush4R2. Maybe this caused it.
I have reinstalled ZBrush 4R2 and it is starting again now.
