I am very new to ZBrush. I would like to design a vase to print on my 3d printer. My problem is, if I start with a cylinder, and set the inner radius, I get a hole all the way through. I need to have a bottom.
What are the units in the init subpallet? I can set a size to 50, but 50 what?

You can use Extract on a whole cylinder.
ZBrush doesn’t use real world measurements. What you see on the sliders in the “Initialize” subpalette are a percentage of the original. For 3d printing purposes, real world measurements can be used in the 3D Print Exporter plugin within ZBrush when you are ready to export. You can find the documentation for 3D Print Exporter in your “ZBrush 4R6/Documentation/plugins” folder.
