I’m a new to ZBrush, and I bought a Wacom Intus3.
I have a spray pen (With a small color wheel)
I want to setup ZBrush as best as I can to have a good workflow…
anyone can help with this?

ZBrush doesn’t need any special setting up for use with a Wacom tablet. There are settings you can change in the Preferences>Tablet menu but I expect most people don’t touch those. Also you can set tablet options per brush if you want - see the Brush>Tablet Pressure menu.
I don’t know about the spray pen but I expect you will find that pressure is the only pen input ZBrush will respond to (apart from mouse type buttons).
