Sculpt and paint with spotlight

So I want to use a photo of my tongue surface to polypaint the dorsal surface of a tongue model. I also have taken this photo and in Photoshop manipulated it into a greyscale photo that more correctly creates the nooks and crannys and tastebuds than the full color photo does when sculpting with spotlight. So are there tricks to using one photo in, I guess spotlight, to polypaint color and then using a different photo to sculpt detail in polypaint but having them be them perfectly aligned? And if you can’t get it right in one try, how can you get back to the same alignment in spotlight to touch it up, or do you just have to eyeball it dest you can?

As I mess with this… It doesn’t really work the way I had expected. Even though the tops of the taste buds are different color the valleys, works better to paint the rgb and manually sculpt the surface details to fit with the polypaint.
