Is there any way I can save the transpose unit settings, which presently I re-calibrate at each Zbrush start up, to either a file or zbrush start-up config? Thank you for the help, and forgive me if this question is asked too often.

You could record a Macro of the changes and have it run automatically at startup.
edit: or you can just move the new Macro button to a convenient spot and click it to make the changes.
