let´s say I positioned, scaled and rotated a low detailed version of the subtool to make sure it fits with the rest of my model and then want to further detail it. Is it possible to replace the subtool? Thank you

What is it that’s keeping you from subdividing it further and detailing it?
errrr, basically nothing, but because I already rotated it I do have problems drawing my masks properly.
You mean, because of the broken symmetry? Alas, there is no way that Im aware of to restore symmetrical operations to an object that has been rotated and positioned asymmetrically in the worldspace. Poseable symmetry, I dont think is applicable here. When constructing a symmetrical object that will be placed asymmetrically in a model, always get the basic form and symmetrical details worked out first.
Beyond that, I dont understand why you would be having trouble painting masks. You can, though, export any subtool into an external application, do any work on it, or replace it with new geometry entirely, then reimport it into Zbrush (but not directly into the active tool), append it as a subtool, and it should occupy the same position as before.
Note, that some people are having issues with scale and position being off when reimporting subtools like this. I recommend using the “Scale Offset” function in subtool master to “zero out” the subtools position prior to export.
Theoretically, you can even import different geometry directly into the base subD level of a subtool now, and Zbrush will update it for you, though I am distrustful of this.
