Yes, this is the way you would apply Dynamesh multiple times. However, keep in mind that if the “Project” button is not enabled in the Dynamesh menu Dynamesh will not make any changes unless the mesh itself has been changed. If no verts on the mesh have changed position since the last Dynamesh nothing will happen.
Switching on the Project button will force Dynamesh to update each time, but there generally isn’t any benefit to applying Dynamesh multiple times in a row if nothing on the mesh has changed.
