I try to change the pen eraser button to behave as click function in ZBrush, but no matter what key or function is assign in Wacom settings does not reflect in ZBrush.
Does anyone face the same problem?

I’m finding the same thing you are. By default ZBrush is set to use the eraser button as a ZSub alternative for the active brush, painting with negative displacement with whatever the active brush is. This behavior appears to override any custom eraser button settings when using the eraser button to sculpt on the mesh. When not over a mesh and not being used to sculpt, custom settings appear to function as expected.
You can, somewhat counter intuitively, force the button to behave like a normal brush by setting the behavior to ZSub. This will invert the brush to positive displacement. However, the normal ALT button behavior that inverts the ZAdd/ZSub behavior does not work with the eraser button.
I’ve notified the development team of this to be look at.
